Julian Redelinghuys has been ruled out for six weeks due to a hamstring injury sustained in South Africa's Rugby Championship opener against Argentina.
The prop did the damage in the first half of Saturday's 30-23 victory over the Pumas at the Mbombela Stadium in Nelspruit.
Sharks tighthead Lourens Adriaanse will travel with the squad to Salta for the return fixture next weekend, while Trevor Nyakane could make his return from an ankle injury.
Springboks head coach Allister Coetzee. said: "Losing Julian is a massive blow to us because it comes at a time when he is playing really good rugby,
"Vincent Koch did extremely well when he came on a replacement and Julian's injury now presents him with an ideal opportunity.
"The players showed great resilience on Saturday, they stayed calm and stuck to the plan, especially in the last 10 minutes.
"We continue to work and focus on our team cohesion and we know the challenge we will face in Argentina."
Lions duo Malcolm Marx and Franco Mostert are set to join up with the South Africa squad for the Salta Test, along with Stade Francais fly-half Morne Steyn.
