Munster's Earls is suspended for two weeks for a dangerous tackle on Glasgow's Fraser Brown, while Jackson has been omitted from the group due to personal reasons.
Forwards Iain Henderson, Sean O'Brien and Peter O'Mahony are injured, while Luke Marshall and James Tracy were also dropped from the initial 34-man squad.
In the absence of Jackson, 21-year-old fly-half Joey Carbery could, if called on, make his Test debut as Jonathan Sexton's understudy.
